I admire your willingness to take the plunge and drive into Manhattan for the first time. A lot of good freight comes out of NYC. By choosing to be one of those who will go in after it, you avail yourself to more opportunities than those who don't.
Aprehension about the unknown is understandable. It will pass as your knowledge of the city grows. You can accelerate the learning curve by purchasing paper maps of the city, detailed enough to show one-way streets, and studying them. It became much easier to get in and out when we bought computer mapping software.

This may not apply to your specific pickup, but not seeing it mentioned here let me share one of our favorites routes into and out of Manhatten. It is not always the shortest but we find it quite easy.
If you are coming in via northbound I-95, and crossing the George Washington bridge, continue to I-87, the Major Degan Expressway. Go South on I-87 to the 3rd Avenue bridge exit. Take the bridge across to Manhattan. Pick up Lexington Ave from there. Lexington is a one-way street that runs as far down as 23rd Street if you need it.
On the map, you will see Madison Ave just two blocks west of Lexington. Madison Ave is one way running north, all the way to the Madison Ave. Bridge, which puts you back on I-87.
Again, using these routes may not be the shortest or even the best. Everyone has the routes they like. Having discovered the Madison Ave bridge, I'm in love with Madison Ave. Once we're on that one-way street, we're as good as out of the city.
The others are correct. Stay off the parkways with a truck. Commercial trucks are not permitted there.
For your pickup, using the above bridges, take I-87 to the 3rd Ave Bridge and pick up Lexington Ave. Follow Lexington to 47th St, which is one-way to the right. Turn right onto 47th St. and follow it to Broadway and your pickup. Note that 48th st and 46th street are one-way coming back the other way. Also note that shortly after you turn right on 47th, you will cross Madison Ave. On your way back out, take either 48th st or 46th st to Madison Ave. Turn Left on Madison Ave and follow it all the way to the Madison Ave. Bridge. You pick up I-87 North just across the bridge. It is a left turn onto the ramp.
Note: Last time we did this, a few months ago, there was construction just off the 3rd Ave bridge. Keep alert and follow the signs through the construction (provided no one has stolen them or covered them with graffitti). It would be wise to do a close map study of the area just beyond the 3rd ave bridge to get you onto Lexington.
